Abstract : Mosquitoes may carry millions of microorganisms that, in large enough doses, can cause disease. Garbage, manure, and similar wastes that cannot be made inaccessible to flies can be treated with larvicidal drenches or dusts. Residual insecticidal sprays are effective against flies for several weeks; however, some houseflies have developed resistance to certain insecticides, such as DDT. More than 100 pathogens associated with the mosquitoes may cause disease in humans and animals. Pathogenic organisms are picked up by flies from different sources, and then transferred on their mouthparts and other body parts, through their vomitus, feces and contaminated external body parts to human and animal food. Biological control play a main role in the recent years for controlling insects so that we will try to develop new genetically modified BT strains for controlling different types of mosquitoes. In the present study a total of 19 Bacillus thuringiensis were isolated from soil samples Jeddah, Saudi Arabia. The distinctive properties of 16s rRNA gene as universal phylogenetic marker was employed to carry out comprehensive diversity study of the bacterial isolates in order to reveal the evolutionary and phylogenetic relationships among these isolates. A random amplified polymorphic DNA (RAPD) and ISSR fingerprinting method has been used to fingerprint isolated 19 Bacillus thuringiensis bacterial strains using 6 different primers. The RAPD profiles revealed a high level of DNA sequence diversity. Random mutation with chemical (UV) and physical mutagenesis (Sodium Azide) were also done on some chosen Bacillus thuringiensis strains. Study of mutants obtained against larva of mosquitoes in comparison with wild type strains. Results showed that some of the obtained mutants are promising in its mortality against larva some mutants give us mortality up to 80% which is excellent against larva.
